Mini-prep Protocol

Protocol

 

 

* Materials

 

- Mini-prep Kit: QIAGEN, spin column

- 세포 배양 적당량: ~10ml

- LB broth

  1) Dissolve LB-Broth (25g/L) in DW and autoclave

  2) Leave the solution at RT to cool down

  3) Add Ampicillin at a concentration of 100ug/ml just before using

 

 

* Methods

 

- Transformation이 완료되어 생긴 colony 하나를 LB broth 에서 O/N 배양 (37℃, shaking)

 

1. 2ml tube에 배양액 1.9m씩 넣어주기

2. Centrifuge 6000 rpm, 1 min, 4℃, supernatant 버리기

3. Centrifuge 되는 동안 P1, P2, P3 buffer 준비

  1) P1 (Resuspension buffer) 250λ 넣고 vortexing, pellet 풀어주기

  2) P2 (Lysis buffer) 250λ 넣고 inverting

  3) P3 (Neutralization buffer) 350λ 넣고 inverting

4. Centrifuge 13,000rpm, 10 min, 4℃

5. Spin column에 supernatant 붓고 Spin down 해서 filter

6. PB buffer 500λ 넣고 spin-down

7. PE buffer 750λ씩 넣고 spin-down

8. 추가적으로 Spin down, PE 완전히 제거

9. 뚜껑 열고 10분간 dry, DNA tube의 filter 부분만 EP tube로 옮겨주기

10. EB (Elution buffer) 50λ 뿌린 뒤 1min 기다리기

11. Centrifuge 13000rpm, 1 min, 4℃

12. Nanodrop으로 purity check
